ESP Science Users meeting

Issues:

  1. (some) probes ordered for JdF.  Some were too clipped in the melt-curve analysis and we're waiting for new sequence from Harvard 
  2. Gordon out of Ball but still with some reagents.  Chris to run some samples collected at diffuse vent field at JdF in 2004 to see if we can get both PCR and SHA from same sample.
  3. Working on PCR paper.

  1. JdF wand design
    1. Spoken with Geoff Wheat who suggests getting into the vent crack.  deeper into crack = higher temperature = more microbes. Geoff has two Antara temp probes he might loan.  Also could get some custom HOBO's, but these wouldn't be real-time.  Scott being consulted....
    2. Roman waiting to hear from Vince @ Seabird.  They are suggesting some options, but unclear whether their temperature gauges can go hot enough.  Currently choices are A.) Seabird, B.) interface our own probe, C.)Adam Schultz--contractor who might make us probes.  Will work with engineering on making this decision.
    3. Some discussion of alternative site with hotter vents.  Currently think that will have max of 40C at Ashes vents.
    4. There will be no sediment at ashes.  It's  ROCK.

  1. Ran experiment looking at array sensitivity of array when lysed from pressure pots vs. on bench.  Both results were similar.  This is bedrock work before going into cold temperature.  Ultimately trying to see if cold reduces our array sensitivity; does a correction factor need to be introduced based on environmental temperature.

  1.   Bruce being reassembled and will perform the room temp tests (full run) end of this week.  Then into cold over weekend, and cold test early next week.
  2. Clamps on MOE would not compress the o-ring, so poor thermal contact on both clamps with pucks.  Mike P looked at and found that grooves were too shallow.  He will examine drawings and fix.  Jim will email this issue to McLane. 

 OTHER ISSUES:

 Chris S. sent email to Bruce K asking what their decision was for this spring deployment.  Still waiting to hear.

Optode:  Two types: Clark is sensitive to oxygen, but also to H2S and needs frequent servicing.  Optode needs far less servicing, but is less sensitive at low oxygen concentrations.  Should we get Seabird to add an optode to one of our CTD's?  Decision was yes, we could use both types.  Will charge to 900915.
